Its main advantage lies in having two similar pKa values of 6.8 and 9.0, respectively, which gives it a very wide buffering range, approximately between pH 6.3 and 9.5. The pH of this buffer can be as low as 6-7, which makes it more suitable than Tris buffer for improving the stability and activity of restriction endonucleases. The application fields of Bis Tris propane are very extensive. In the study of DNA polymerase fidelity and PCR reactions, it has been used as a buffer with good results.
